    • Bouchon A., Facchetti F., Weigand M.A., Colonna M. TREM-1 amplifies inflammation and is a crucial mediator of septic shock. Nature. 410:2001;1103-1107. Bouchon et al. have examined the role of TREM-1 - a new receptor of the immunoglobulin superfamily that is expressed on neutrophils and monocytes. Upregulation of this receptor is associated with bacterial infections in humans, and studies show that TREM-1 is an amplifier of microbial induced inflammatory responses.

In this study, transgenic mice were made expressing β2-microglobulin on DCs using the CD11c promoter. The CD11c-β2-microglobulin transgene was bred onto a β2-microglobulin-deficient background. Using bone marrow chimeras (donor CD11c-β2min) made with mice expressing the RIP-ova transgene, the authors could demonstrate that the reconstituted expression of class I molecules on DCs was sufficient to promote cross presentation and tolerance in vivo.

Nature. 416:2002;603-607. B cells capable of making rheumatoid factor autoantibodies were stimulated in vitro by addition of anti-chromatin antibodies. Activation of the B cells occurred via a MyD88-dependent signalling pathway and had other properties suggesting it may involve a TLR, perhaps TLR9. These studies suggest that innate immune receptors may play a key role in the breakdown of B cell tolerance in systemic autoimmunity.

Immunity. 15:2001;775-785. Evidence is presented that Sle1c represents a partial loss of function allele of Cr2, which encodes the complement receptors CR1 and CR2 in the mouse. Although previous work has indicated that CR2 is a positive factor for B cell activation, there was also evidence it participates in induction of tolerance of B cells. The latter function could explain how a partial loss of CR2 function contributes to the development of autoimmunity.

Competition for BLyS-mediated signaling through Bcmd/BR3 regulates peripheral B lymphocyte numbers. Curr Biol. 11:2001;1986-1989. A demonstration that A/WySnJ B cells cannot respond to BAFF. This work demonstrates that the BAFF receptor is the key receptor mediating the in vivo survival effect of BAFF on B cells.

CD28-dependent activation of protein kinase B/Akt blocks Fas-mediated apoptosis by preventing death-inducing signaling complex assembly. J Exp Med. 196:2002;335-348. This paper demonstrates that active PKB is able to impair the Fas death signalling pathway by inhibiting the recruitment of procaspase 8 to the death inducing signalling complex.
