Cytotoxic cell granule-mediated apoptosis: Perforin delivers granzyme B-serglycin complexes into target cells without plasma membrane pore formation
-
This paper makes two intriguing observations: firstly that a proportion of cells exposed to perforin and granzyme B undergo cell death without overt plasma membrane pore formation; secondly, that granzyme B and perforin may be co-delivered to a target cell on a scaffold of cytotoxic granule-derived proteoglycan to which they are held by electrostatic forces.

A clathrin/dynamin- and mannose-6-phosphate receptor-independent pathway for granzyme B-induced cell death
-
Granzyme B enters cells through two independent endocytic mechanisms, one requiring cell-surface receptor binding and the second occurring through constitutive fluid phase uptake. In the absence of receptor-mediated uptake, fluid phase uptake can still efficiently deliver granzyme B for the purposes of apoptosis induction.

Caspase activation by granzyme B is indirect, and caspase autoprocessing requires the release of proapoptotic mitochondrial factors
-
This paper defines the downstream events in caspase activation in response granzyme B. The authors identify several pro-apoptotic factors released from mitochondria that are crucial for displacing IAPs from partly processed caspases. This enables caspase activity to reach lethal threshold in targeted cells.
