Large meta-analysis establishes the ACE insertion-deletion polymorphism as a marker of Alzheimer's disease
-
Lehmann DJ, Cortina-Borja M, Warden DR, et al. Large meta-analysis establishes the ACE insertion-deletion polymorphism as a marker of Alzheimer's disease. Am J Epidemiol 2005; 162:305-317. After analysing data from 6037 patients with Alzheimer disease and 12 099 controls, the authors conclude that angiotensin I-converting enzyme (ACE) indel is a marker for the risk of Alzheimer disease, probably in linkage disequilibrium with the true risk polymorphism.

Schliebs R. Basal forebrain cholinergic dysfunction in Alzheimer's disease interrelationship with beta-amyloid, inflammation and neurotrophin signaling. Neurochem Res 2005; 30:895-908. The author reviews the complex interrelationship between cholinergic neurotransmission, APP metabolism and -amyloid-induced production of proinflammatory cytokines hypothesizing that an age-related disturbance of the homeostatic interaction between cholinergic function and APP metabolism plays a major role in the genesis of SAD.

Decreased adult hippocampal neurogenesis in the PDAPP mouse model of Alzheimer's disease
-
Donovan M, Yazdani U, Norris R, et al. Decreased adult hippocampal neurogenesis in the PDAPP mouse model of Alzheimer's disease. J Comp Neurol 2006; 495:70-83. While some studies link Alzheimer disease to increased neurogenesis, in the PDAPP the primary characteristic is decreased neurogenesis. This feature might contribute to the cognitive decline and thus could be used as a supplementary means to evaluate Alzheimer disease therapies.

Treatment with an amyloid-beta antibody ameliorates plaque load, learning deficits and hippocampal long-term potentiation in a mouse model of Alzheimer's disease
-
Hartman R, Izumi Y, Bales K, et al. Treatment with an amyloid-beta antibody ameliorates plaque load, learning deficits and hippocampal long-term potentiation in a mouse model of Alzheimer's disease. J Neurosci 2005; 25:6213-6220. PDAPP mice, on a pure C57BL/6 background, tested on a 5-week water maze protocol, have early learning impairments and exhibit greater cognitive deterioration with age than their WT littermates. The authors also show that after systemic treatment for several weeks with an anti-Abeta antibody, the performance of animals with high Abeta burden is improved.

Proc Natl Acad Sci USA 2005; 102:14854-14859. Environmental enrichment reduces Abeta levels and amyloid deposition in transgenic mice
-
Lazarov O, Robinson J, Tang Y, et al. Environmental enrichment reduces Abeta levels and amyloid deposition in transgenic mice. Cell 2005; 120:701-713. The study highlights the efficacy of environmental enrichment in reducing levels of brain Abeta and amyloid depositions in transgenic mice. The authors also present data showing that physical exercise lowers brain Abeta levels. These findings are intriguing in light of epidemiological data linking the risk for development of dementia to environmental factors such as education and life style.
