    • This study defines sequential requirements for the initiation and propagation of regional heterochromatic domains. The authors show that the RNAi machinery is required for heterochromatin formation in fission yeast. Genetic and biochemical analysis show that a DNA element with homology to centromeric repeats and RNAi machinery cooperate to nucleate heterochromatin at the silent mating-type interval of fission yeast but are dispensable for its subsequent inheritance. Moreover, histone H3 Lys9 methylation and heterochromatin assembly is nucleated at the specific regulatory repeat region, which then spreads to neighboring sequences in a Swi6/HP1-dependent manner, leading to epigenetic silencing of donor mating-type genes.
