    • Although the conventional view is that members with the TNF-R family exist as independently diffusing monomeric species and trimerize upon ligand binding, some evidence suggests however, that several of these receptors may exist as pre-assembled oligomers prior to ligand binding. It has been shown that a conserved domain in the extracellular region of TNF-R, Trail receptor 1, and CD40, distal from the domain that interacts with the ligand, serves as a preligand-binding assembly domain PLAD, Chan, F.
