Cancer. 2:2002;331-341 A comprehensive discussion of the molecular mechanisms of the multistep process of tumorigenesis in humans and mice. This review concisely summarizes the complex body of experimental data on the process of malignant transformation and discusses the discrete histopathological steps and their correlation with specific genetic mutations. The authors also discuss the strengths and weaknesses of rodent models of human cancer and several of the important signaling pathways that differ between these species. Discussion of telomere shortening and other barriers to cellular immortalization are discussed in the context of cell culture as well as in vivo model systems.
