The authors conclude that THO/TREX may prevent the formation of DNA:RNA hybrids during elongation by promoting efficient mRNP assembly.

The individual subunits, Snu17p, Bud13p and Pml1p, are not essential for viability but are required for efficient splicing in vitro and in vivo. Inactivation of this complex causes pre-mRNA leakage from the nucleus. In particular, deletion of Pml1p results in significant mRNA leakage, whereas splicing defects were only mild, indicating that the major function of Pml1p is to promote the nuclear retention of unspliced mRNA.

Importantly, Mlp1p is required for this quality control mechanism without being implicated in splicing itself. Moreover, it is shown that Mlp proteins are asymmetrically distributed around the nuclear envelope, being only found in regions abutted by chromatin.

Dbp5p associates physically with transcription factor IIH components and these interactions affect not only growth but also mRNA export, indicating a role for Dbp5p in both, transcription and mRNA export.
