    • A nonredundant role for the adapter protein Shc in thymic T cell development
    • The authors show that Shc is tyrosine phosphorylated after pre-TCR engagement in immature DN thymocytes. To further demonstrate the in vivo role of Shc in thymocyte development the authors use two genetic approaches. First, by conditional inactivation of the Shc1 genomic locus it is shown that Shc is required to transduce signals required for the transition from the DN3 to the DN4 stage through the pre-TCR. To further assess the role of Shc in T-cell development, the authors generated transgenic mice expressing a dominant-negative form of Shc in which the tyrosine residues Tyr239, Tyr240 and Tyr317 were mutated to phenylalanine. Similar to the conditional deletion, the expression of the dominant-negative Shc also results in a partial developmental block of DN thymocytes. The study thus demonstrates that Shc and signalling pathway(s) downstream of Shc are essential for optimal pre-TCR signalling and progression through the DN stage.

    • A mouse with a loss-of-function mutation in the c-Cbl TKB domain shows perturbed thymocyte signaling without enhancing the activity of the ZAP-70 tyrosine kinase
    • By producing mice with a knock-in mutation (Gly304→Glu) in the TKB domain of c-Cbl, which abolishes the interaction with phosphorylated Tyr292 of ZAP-70, the authors investigate the functional requirement of the TKB domain in TCR-mediated signalling events in thymocytes. Conversely to c-Cbl-deficient mice, Gly304→Glu knock-in mice show normal surface levels of the TCR-CD3 complex and normal ZAP-70 activity. In contradiction to previous models, Thien and co-workers here show that the TKB domain is neither required to negatively regulate ZAP-70 activity nor to regulate TCR-CD3 expression in immature thymocytes.

    • In this paper the authors investigate the mechanisms underlying the previously described B-cell defect in BCAP-deficient mice. They find that BCAP determinates B-cell longevity by regulating c-Rel expression, and the induction of pro-survival molecules and factors such as cyclin D2 and Cdk4, which are required for cell-cycle progression.

    • This study uses transgenic mice overexpressing PAG to demonstrate the inhibitory effect of PAG upon T-cell activation. The inhibitory effect is alleviated in the Tyr314→Phe mutant, which abolishes the Csk binding site. Interestingly, this mutant appears to enhance TCR stimulation, suggesting that PAG may also possess positive regulatory activity.

    • The authors demonstrate that Cbl-b deficiency in T cells does not affect the stability and the expression of CRK-L, but rather its association with C3G. The enhanced interaction between CRK-L and C3G increases activation of the GTPase Rap1 to promote adhesion to LFA-1 in Cbl-b-deficient T cells. This suggests that Cbl-b plays a negative role in CRK-L-C3G-mediated Rap1 and β2-integrin activation in T cells.
