    • Synthetic anti-BR3 antibodies that mimic BAFF binding and target both human and murine B cells
    • Shotgun alanine scanning was used to explore the mechanisms whereby a synthetic antibody originally raised against mouse BR3 was evolved to cross-react with human BR3. It was revealed that cross-species binding was achieved using a common hot spot that interacted with conserved residues in the two antigens, and subsequently by accumulating mutations that enabled better accommodation of human BR3.

Notably, phage display failed in this system and, thus, this method might be useful for proteins that are recalcitrant to phage display.
