Evidence that the anti-inflammatory effect of LXR is mediated by inhibition of nuclear factor-κB activation is shown.

This paper demonstrates that LXRα knockout mice on a cholesterol-containing diet fail to upregulate hepatic Cyp7a expression, resulting in massive cholesterol accumulation in the liver, impaired hepatic function and lethality. Failure of LXRβ to compensate for LXRα and normal phenotype of LXRβ null mice highlights distinct roles of hepatic LXRs.

Science (2000) 289(5484):1524-1529. This paper reports that treatment of mice with T-0901317 decreases cholesterol absorption. This is attributed to upregulated ABCA1 expression in small intestine, however, subsequent discoveries (see references [5-9]) indicate that this effect is due to upregulated intestinal ABCG5 and ABCG8 expression. Studies in peritoneal macrophages from knockout mice demonstrate upregulation of ABCA1 mediated by both LXR subtypes.

Studies in cholesterol fed and T-0901317-treated wild-type, LXRα- and LXRβ-deficient mice show that hepatic LPL is upregulated preferentially by LXRα. LPL upregulation is mediated by LXRE in intron 1, which shows higher activity with LXRα versus LXRβ in gel shift and transient transfection experiments.
