A beautiful and elegant study of the dynamic localisation of green fluorescent protein tagged PAR-2 and PAR-6 in live embryos. The authors show that the complementary asymmetric localisation of these proteins is achieved in distinct initiation and maintenance phases. The first phase depends on sperm astral microtubule contact with the cortex, and the second involves mutual exclusion of PAR-2 and the PAR-3-PAR-6-PKC-3 complex.

This paper provides new data on Drosophila PAR-1 in oocyte polarisation. The authors raised an antibody to the amino terminus of the N1S isoform of PAR-1 and found that it detects a different pattern of localisation than previously raised pan-PAR-1 antibodies [31,32]. The N1S antibody detects PAR-1 at the posterior of early oocytes (the pan-PAR-1 antibody shows PAR-1 on the fusome and at the posterior of later oocytes), and a complementary localisation of Baz/PAR-3 and N1S PAR-1 is seen, providing a further parallel between Drosophila oocytes and the one-cell C. elegans embryo. A model is presented whereby the fusome, microtubules and Baz/PAR-3 enrich and localise PAR-1 to the posterior of oocyte, where it functions to polarise the microtubule cytoskeleton.

The maternal gene spn-4 encodes a predicted RRM protein required for mitotic spindle orientation and cell fate patterning in early C. elegans embryos

This study of C. elegans spn-4 mutants showed that the mitotic spindle is polarised and can localise cortical and cytoplasmic factors to one pole. A similar activity of the spindle is seen in Drosophila neuroblasts [87]. SPN-4 encodes a protein with an RRM domain (putative RNA-binding). Besides the defect in spindle orientation, spn-4 mutants also have defects in other processes such as specification of mesendoderm and germline quiescence during embryogenesis. SPN-4 is proposed to post-transcriptionally regulate the expression of a variety of targets, none of which are yet known.
