Science 1997, 278:2085-2092. This paper is more remarkable for its brilliant chemistry than for its cellular immunology, but by introducing 'reactive immunization' - that is, immunizing with antigens that react with the combining site of certain antibodies arising by somatic mutation - Lerner and co-workers may have provided us with a wonderful tool for studying the selection of mutants with higher affinity in germinal centers.

J Exp Med 1998, 188:1453-1464. These two papers [29•,30•] from the laboratory of David Nemazee - the father of 'receptor editing' - report that antibodies of low affinity, but not those of high affinity, induce RAG re-expression in germinal center B cells. Now that finding just has to be important - regardless of whether RAG is re-expressed because of, or despite, the fact that it can result in V-segment replacement.

    • Mismatch repair co opted by hypermutation
    • Cascalho M, Wong J, Steinberg C, Wabl M: Mismatch repair co opted by hypermutation. Science 1998, 279:1207-1210. Our paper was the first to report that knocking out a DNA-repair gene affected hypermutation. It makes the still-controversial proposal that mutations in hypermutable sequences are fixed by the components of the Pms2/Msh2 repair pathway.

    • Rada C, Ehrenstein MR, Neuberger MS, Milstein C: Hot spot focusing of somatic hypermutation in MSH2-deficient mice suggests two stages of mutational targeting. Immunity 1998, 9:135-141. This important paper butresses the notion that hypermutation depends on DNA repair, by finding a change in the mutational spectrum as well as a reduced frequency of mutations when the Pms2/Msh2 pathway is rendered nonfunctional.
