    • Precursor B cells showing H chain allelic inclusion display allelic exclusion at the level of pre-B cell receptor surface expression
    • -/- mice were analyzed for their heavy-chain rearrangement status. In both mice, a small proportion of cells (3-7%) was found to have both heavy chain alleles rearranged; one of these alleles encoded a μH chain that could form a pre-BCR while the other μH could not. These findings imply that allelic exclusion is induced by a pre-BCR signal, but does not require λ5.

This paper reports the effects on B cell development and allelic exclusion of an immunoglobulin-transgene specific for phosphorylcholine, in which the ability of the μ heavy chain to associate with CD79a-CD79b is compromised (by a YS→VV mutation). B cell development is blocked at the pre-B cell stage, while allelic exclusion is maintained. This demonstrates that CD79a-CD79b-mediated signaling is required for B cell development to pre-B and immature B cells and suggests that allelic exclusion may be induced via a different pathway.

    • Epstein-Barr virus LMP2A drives B cell development and survival in the absence of normal B cell receptor signals
    • + cells exhibit no um rearrangements but pre-B cells do contain light-chain rearrangements. LMP2A thus mimics signals that are normally mediated through the CD79a-CD79b-calnexin complex and the pre-BCR. Rearrangement of μm does not occur because LMP2A is expressed before the pre-BCR is assembled and provides an alternative signal.

Mice transgenic only for the immunoglobulin heavy chain were generated and pairing with the endogenous light chains led either to B cells which were specific for Sm or single-strand DNA or to non-autoreactive B cells. Autoreactive B cells are eliminated at the level of the transitional B cell. The authors propose that this late tolerance induction is due to a combination of low affinity BCRs and low antigen concentration and that elimination occurs as a result of increased mIgM expression by transitional B cells.

SLP-65 shows homology to SLP-76 which serves as an important linker molecule in T cells. In B cells, SLP-65 is rapidly tyrosyl-phosphorylated by Syk upon BCR aggregation - providing SH2-dependenl binding of PLCγ, Vav, Grb2 and Nck. The central role of SLP-65 in BCR-mediated signaling is illustrated by its positive effect on downstream calcium mobilization.

In [29•,30•], the role of signaling intermediaries in BCR-mediated activation of MAPK family members p38, JNK and ERK are investigated in the chicken lymphoma model DT40. ERK activation depends on Ras, PLCγ and Syk but not on Lyn or Shc; JNK activation depends on Syk, Btk, Ras and PLCγ but not on Lyn; p38 activation depends on Ras and PLCγ but is intact in Lyn-, Btk- or Syk-deficient DT40 cells while the absence of both Lyn and Syk inhibits p38 activation. Together, the data demonstrate that, at least in this model, BCR-mediated MAPK activation crucially depends on PLCγ and Ras while activation is differentially regulated at the level of protein tyrosine kinases.

This is an important contribution, investigating the role of BCR expression in mature B cells using inducible-gene-targeting techniques. Ablation of BCR expression led to rapid cell death, preceded by downregulation of MHC class II and upregulation of CD95 (Fas). Cell death is delayed by constitutive bcl-2 expression, supporting the notion that cells die by apoptosis. Together the data indicate that continued B cell survival in the periphery actively depends on BCR expression.

These findings help to define regulatory steps in early B cell development.

In the absence of tyrosyl phosphorylation, CD22 does not recruit SHP-1 - which is necessary for the negative regulation of calcium mobilization. In addition, all three molecules appear to be limiting in this negative regulatory pathway.

As demonstrated, SLP-76 is expressed in T lymphocytes, macrophages and natural killer (NK) cells but is absent in B lymphocytes. Genetic ablation of SLP-76 leads to an early block in T cell development and there are no mature T cells present. Despite expression in normal NK cells and macrophages, these compartments are intact in knockout mice and - in line with the expression pattern - no B cell abnormalities were observed. Thus SLP-76 is crucial in mediating TCR signals early in development.

NF-κB and JNK are selectively activated by a large, transient rise in intracellular calcium whereas NFAT is activated by a low, sustained calcium plateau; furthermore, BCR-mediated signaling in naive cells triggers a large biphasic calcium response whereas low calcium oscillations are observed in tolerant B cells. Together these data reveal a mechanism by which a multifunctional second messenger, such as calcium, can achieve specificity in signaling to the nucleus and demonstrate the biologic significance of these events in naive and tolerant B cells.
