In this paper, the authors demonstrate the important role of electrostatic transition state stabilization in the chorismate mutase catalyzed chorismate rearrangement. Two amino acid positions near the active site were randomized. A positive charge at either position was shown to be essential for catalysis. This analysis led the authors to postulate that the electrostatic nature of the transition state in the enzyme-catalyzed reaction was similar to that in the uncatalyzed reaction.

In this study, the dependence of distance between the two catalytic glutamates of the enzyme xylanase was explored through site-directed mutagenesis and the introduction of non-natural amino acid sidechains. Lengthening the distance through a mutation of the nucleophilic glutamate to aspartate as well as shortening the distance through mutation of the glutamate to a cysteine (which was then treated with iodoacetate to give the carboxymethylated cysteine) led to decreased xylan hydrolysis.

The authors demonstrate that the variant GFPs are capable of fluorescence resonance energy transfer, thereby making them a useful tool for monitoring protein-protein interactions.
