    • Pasare C, Medzhitov R. Toll pathway-dependent blockade of CD4+CD25+ T cell-mediated suppression by dendritic cells. Science 2003; 299:1033-1036. This important study examines the interaction between the TLR pathway with T regulatory cells via the activation of dendritic cells. The stimulation of TLR induces the production of IL-6, which in turn blocks the activation of T regulatory cells allowing activation of pathogen-specific adaptive immune responses. This blockade could thus prevent chronic infection and autoimmune diseases.

This study demonstrates that MyD88-deficient dendritic cells can prime naive T lymphocytes towards Th2 responses. MyD88 seems not to be involved in the modulation of the Th1/Th2 balance. A MyD88 independent pathway may play a role in allergic immune responses.

This study shows a decrease of Th2 responses in TLR4-deficient mice sensitized to inhaled allergen. The expression of co-stimulatory molecules and cytokine production are decreased in TLR4-deficient dendritic cells and may explain the Th2 priming impairment.

Low-dose inhaled lipopolysaccharide combined with allergen induces Th2 responses, whereas high-dose lipopolysaccharide resulted in Th1 responses.

The immunostimulatory DNA could be useful in the treatment of human inflammatory bowel disease.

Oral administration of λ-carrageenan to immunized mice decreases OVA (ovalbumin)-specific IgE and is also effective in previously immunized mice. This effect is dependent on a TLR4 and MyD88 pathway with increased IFN-γ and decreased IL-4.
