Cell 1997, 90:785-795. Appearing contemporaneously with the study of Nakamura et al. (Science 1997, 277:955-959), this article identified the human gene encoding the catalytic subunit of telomerase and named it hEST2. Expression of this gene, which was mapped to chromosome 5p15.33, was noted at high levels in primary tumors, cell lines, and telomerase-positive tissues but was undetectable in telomerase-negative cells and tissues. The Human Genome Organization Nomenclature Committee renamed this gene hTERT.

This article reports the cloning of the human gene that encodes the catalytic subunit of telomerase. Named hTRT, this gene was identified on the basis of its homology to telomerase-encoding genes in Saccharomyces cerevisiae and Euplotes aediculatus. Sequence comparisons placed the telomerase proteins in the reverse transcriptase family. Expression of hTRT correlated with the presence of telomerase activity in cell lines, suggesting that telomerase activity is regulated, in part, by the transcription or stabilization of nTRT mRNA.

The product of the EST2 gene was identified as a subunit of telomerase in the yeast S. cerevisiae. Immunochemical experiments showed that the EST2 protein is an integral subunit of the telomerase enzyme complex. Mutations in EST2 resulted in the absence of telomerase activity, suggesting a direct catalytic role for the EST2 protein.

When an epitope-tagged version of hTERT was ectopically expressed in telomerase-negative cells, telomerase activity was induced with levels comparable to those seen in immortal telomerase-positive cells. The authors concluded that the synthesis of hTERT protein subunits represents the rate-limiting determinant of cellular telomerase activity.

Telomerase-negative normal fibroblasts were transfected with vectors encoding wild-type and mutant forms of hTERT. Expression of wild-type hTERT resulted in detectable telomerase activity, whereas expression of hTERT mutants did not. Hepatocellular carcinoma and noncancerous liver tissues were evaluated for telomerase activity and hTERT, hTR, and TP1 expression. Only hTERT correlated with telomerase activity, suggesting that expression or stabilization of hTERT transcripts plays a key role in the regulation of telomerase.

Mutations in the reverse transcriptase motifs reduced or abolished enzymatic activity. Normal human fibroblasts transiently expressing hTERT turned on telomerase, suggesting that hTERT is the limiting component necessary for the activation of this enzyme.

Two telomerase-negative human cell types, retinal pigment epithelial cells and foreskin fibroblasts, were transfected with vectors encoding hTERT. In contrast to telomerase-negative control cells, which exhibited telomere shortening and senescence, telomerase-expressing cells had elongated telomeres and divided vigorously without genetic instability for more than 20 doublings beyond their expected life span. This experiment establishes a causal relationship between telomere shortening and in vitro cellular senescence.

    • Telomere shortening and tumor formation by mouse cells lacking telomerase RNA
    • -/- mice lacked detectable telomerase activity, yet were viable for the six generations analyzed. Telomeres shortened over time and were completely eroded by the fourth generation, resulting in aneuploidy and other chromosomal abnormalities. Telomerase-deficient cells could be immortalized in culture, could be transformed by viral oncogenes, and were able to generate tumors in nude mice. The authors conclude that although telomerase is necessary for telomere maintenance, it is not required for oncogenic transformation or tumor formation in mice. These findings might not apply to human malignant transformation; because mouse telomeres are significantly longer than human telomeres, humans may require the activation of telomerase at an earlier stage in the multi-step process of tumorigenesis.

Nat Med 1997, 3:1271-1274. The vast majority of human tumor-derived cell lines exhibit telomerase activity. In this study, 4 of 47 human tumor-derived cell lines lacked telomerase activity but possessed long telomeres. The terminal restriction fragment pattern of these cells differed from that of telomerase-positive cells, suggesting that an alternative mechanism of telomere lengthening (ALT) occurs. Additionally, 4 of 57 tissue samples, representing a variety of human tumor types, had abnormally long telomeres in the face of telomerase negativity. Of the eight tumor samples and cell lines displaying ALT, four were from osteosarcoma. The authors suggest that telomerase is more stringently regulated in nonepithelial cells, which selects for ALT activation as a telomere-lengthening mechanism. The presence of ALT has implications regarding the therapeutic uses of telomerase inhibitors.

A point of controversy is whether telomerase activity is indicative of malignant transformation or is simpiy a marker of active proliferation. In this study, biopsies were performed on normal human uroepithelial cells, and these cells were found to lack telomerase activity. When the same cells were established as proliferating cultures, however, telomerase activity was readily detected. In nonproliferating human uroepithelial cell cultures, telomerase activity was low or undetectable. The authors conclude that proliferative index accounts for the differences between telomerase activity in normal and tumor cells.

Because telomerase is essential for stabilizing DNA, this study tested the hypothesis that inhibition of telomerase would render cells more sensitive to DNA-damaging agents. Telomerase-positive cells from a human glioblastoma cell line were transfected with an antisense vector for hTR, the RNA template of telomerase. This action resulted in inhibition of telomerase and increased susceptibility to apoptotic cell death induced by the agent cisplatin. The authors suggest that inhibition of telomerase may represent a new chemosensitization technique for tumors resistant to anticancer therapy.

Slimb mutant cells accumulated high levels of cubitus interruptus (ci) protein, although transcription of the ci gene was unchanged. Ci was found predominantly in its full-length form in slimb mutant cells, suggesting that slimb, similar to PKA, enhances ci proteolysis and thus antagonizes the hedgehog (hh) pathway.

Notably the mice did not develop BCC, suggesting that BCC, and not medulloblastoma, requires somatic inactivation of the second Ptc allele.

Activating somatic missense mutations in SMO were identified in sporadic BCCs from 3 of 47 patients. Mutant SMO, in contrast to the wild-type, was able to transform rat embryo fibroblasts with the adenovirus E1A oncogene. Transgenic mice overexpressing mutant SMO developed skin lesions with microscopic features similar to BCC. These findings provide direct evidence that mutated SMO can function as an oncogene in the development of BCCs.

Two BCCs demonstrated mutations in both genes. Most tumors showed increased expression of SMO, PTCH, and GLI1 transcripts compared with normal tissue. Thus, in addition to PTCH, SMO mutations may contribute to human tumorigenesis.
