    • Rap1 mediates sustained MAP kinase activation induced by nerve growth factor
    • The authors identified a Rap1-dependent component of NGF signalling to ERKs in PC12 cells. This component accounts for the sustained activation of ERKs following NGF treatment. Moreover, the authors found that an interfering mutant of Rap1 disrupted two features of neuronal differentiation: the expression of selected NGF-responsive genes and the induction of sodium currents. They also identified the Crk family of adaptor molecules as key intermediates for NGF signalling to ERKs

    • Activation of the Drosophila C3G leads to cell fate changes and overproliferation during development, mediated by the RAS-MAPK pathway and RAP1
    • The authors report that overexpression of a membrane-targeted cDNA encoding Drosophila C3G (DC3G) leads to defects in neuronal determination that mimick overexpression of both Rap1 and components of the Ras/ERK pathway. Both loss-of-function mutations of Rap1 and of Ras suppressed the DC3G phenotype, and overexpression of an interfering mutant of DC3G mimicked reduction in ERK signalling. These findings provide the first genetic evidence that C3G may activate ERKs via Rap1, and identify a neuronal function for C3G

J Neurosci. 19:1999;3535-3544. This paper demonstrates that spatial learning in the rat Morris water maze test leads to an activation of ERKs specifically in the CA1/CA2 pyramidal neurons of the dorsal hippocampus. Pharmacological inhibition of MEK did not effect the acquisition, but inhibited the formation, of long-term spatial memory.

    • The MAPK cascade is required for mammalian associative learning
    • The authors found that fear conditioning in rodents elicits NMDA-receptor-dependent ERK activation in the hippocampus. Using a selective inhibitor of MEK, ERK activation was shown to be required for both contextual and cue fear conditioning. This inhibition of associative learning correlated with an inhibition of both ERK activation and the induction of LTP, but not short-term potentiation, in the hippocampus. These findings along with those of Brambilla et al. [44] support a model for LTP representing a physiological phenomenon underlying mammalian learning and memory, and they implicate Ras/ERKs as key regulators of the molecular mechanisms mediating this response
