When neonatal female (C57BL/6 × A/J)F1 mice are injected with an ovarian (ZP3) peptide, autoimmune ovarian disease (OAD) develops at 5 weeks of age. The presence of endogenous antigen at the time of initial priming is essential for the development of a pathogenic autoimmune response as male mice injected neonatally with the same peptide become tolerant to it, and this tolerant state is an antigen specific Th2-deviated response. The autoimmune response (to subsequent ovarian grafts) following neonatal injection is abrogated in female mice that have been neonatally ovarectomised and their T-cell response becomes Th2 dominant. The critical time window ovarectomy effects supports the likelihood that endogenous ovarian antigens are not ignored but are normally perceived by the neonatal immune system.

Transgenic NOD mice that express IL-4 in the pancreatic islets under the control of the human insulin promoter are completely protected from the development of insulitis and diabetes. These mice are not susceptible to disease induction via adoptive transfer of diabetogenic T cells, but islet expression of IL-4 was unable to prevent islet rejection in overtly diabetic recipients. These findings suggest an effect of IL-4 on the early stages of the development of autoimmunity in NOD mice.

This study used a transgenic model in which OVA-specific transgenic CD8+ T cells were injected into unirradiated transgenic RIP-mOVA mice, which express a membrane form of OVA (mOVA) in pancreatic islet β cells and in renal proximal tubular cells, the thymus and in the testis. The findings indicate an extrathymic mechanism that can induce the deletion of CD8+ cells responding to autoantigen expressed in tissues outside the lymphoid compartment. Self antigens are presented, not by the tissues themselves, but by professional APCs in the context of MHC class I via a constitutive exogenous cross-over processing pathway.

This study investigates the diversification of T-cell responses to Bhsp65 in relation to the development of arthritis in susceptible (Lewis) and resistant (WKY) rodent strains with the same MHC class II molecules. Diversification of the T-cell response to the carboxy-terminal determinants of Bhsp65 appears to be involved in protection from autoimmune arthritis. Importantly, the natural trend of diversification in the resistant strain can be approached therapeutically in the susceptible strain via immunization with peptides comprising the carboxy-terminal region of Bhsp65.

J Exp Med 1997, 186:385-391. This paper highlights the importance of IFN-γ in the development of humoral autoimmunity. Antibody response to AChR in IFN-γ KO mice were not demonstrated but an IgG1 response was detected when immunised with a foreign antigen (KLH). The investigators suggest that the difference in response to self-antigen and foreign antigen is due to differences in T-cell affinities. This may result in differential requirements for IFN-γ in the stimulation of B cells for differentiation and subsequent antibody production during cognitive T-B cell interactions.
