    • note
    • Lysosome accumulation ratios (LAR) was calculated in silico using the following equation: LAR = 1 + Σ n = 1 4 Σ i = 1 n 10 p K a i-pH L 1 + Σ n = 1 4 Σ i = 1 n 10 p K a i-pH 0 where pHL is the pH inside the lysosome (assumed to be pH 5.0) and pH0 in the cytosol (assumed to be pH 7.4). This equation is a derivation of the Henderson-Hasselbach equation, based on predicted values of drug p K a according to previous work of Hawley et al.(35) Values of p K a were calculated using ACD/p K a DB software from Advanced Chemistry Development Inc., Toronto, Canada.
