    • In a systematic analysis of the human interactome, a protein matrix of 4456 baits and 5632 preys was screened and 3186 interactions were identified. Subsequent data analysis and validation revealed 911 high-confidence interactions among 401 proteins.

    • A protein-protein interaction network for human inherited ataxias and disorders of Purkinje cell degeneration
    • A comprehensive interaction screening was undertaken for 20 proteins that are involved in inherited ataxias, a class of neurodegenerative disorders with common clinical and pathological features. This work provides an excellent insight into how disease-centered YTH interactome studies can elucidate pathogenic mechanisms and identify candidate genes for complex human diseases.

    • Monitoring protein-protein interactions between the mammalian integral membrane transporters and PDZ-interacting partners using a modified split-ubiquitin membrane yeast two-hybrid system
    • This approach addresses proteins containing PDZ domains that bind to PDZ binding motifs in the C-terminal tails of integral membrane proteins using the MYTH screening system. In this application of the MYTH the transcription factor moiety was shifted to the N-terminus to permit detection of C-terminal PDZ domain interactions.

    • This paper describes the application of transfected cell arrays for mammalian two-hybrid screens. The arrays contain immobilized bait and prey expression plasmids and an auto-fluorescent reporter construct. Adherent cells growing in the array pick the plasmids up and will become fluorescent if the expressed proteins interact.

    • In a first one-hybrid step, informative mutants are selected from randomly generated mutant alleles of one interacting partner (prey). The prey construct contains an additional DBD at its C-terminus, and therefore only full-length proteins can activate the HIS3 reporter gene and grow. In the second two-hybrid step, the selected alleles are tested for interaction with the bait protein by β-gelactosidase expression. Alleles that grow on -His medium not expressing β-galactosidase selectively abolish the bait-prey interaction.
