    • NMR structure of the first PHD finger of autoimmune regulator protein (AIRE1). Insights into autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) disease
    • This article describes the crystal structure of the PHD1 domain of AIRE and confirms its structural similarity to a RING finger domain. Disease-associated point mutations in the PHD1 domain compromise the protein fold responsible for zinc binding, thereby disrupting AIRE function. Interestingly, this group shows that the PHD1 domain is not capable of autoubiquitination.

    • A RING-type ubiquitin ligase family member required to repress follicular helper T cells and autoimmunity
    • The E3 ligase Roquin was isolated in this systematic screen of the mouse genome designed to identify novel single genes involved in autoimmunity. In the absence of Roquin, mRNAs for ICOS and RANTES (among other molecules) are upregulated. It is proposed that Roquin binds to RNA-protein complexes to alter post-transcriptional protein expression, representing a unique method of E3 ligase-mediated regulation.

The E3 ligase Roquin was isolated in this systematic screen of the mouse genome designed to identify novel single genes involved in autoimmunity. In the absence of Roquin, mRNAs for ICOS and RANTES (among other molecules) are upregulated. It is proposed that Roquin binds to RNA-protein complexes to alter post-transcriptional protein expression, representing a unique method of E3 ligase-mediated regulation.
