+ effector T cells develop via a lineage distinct from the T helper type 1 and 2 lineages
-
This study reported inhibitory effects of IFNγ and IL-4 on the development of IL-17-producing effectors. It also demonstrated that STAT1, T-bet, STAT4 and STAT6 were dispensable for development of IL-17-producing effectors in vitro, establishing an independent developmental lineage for Th17.

+ effector T cells develop via a lineage distinct from the T helper type 1 and 2 lineages. Nat Immunol 6 (2005) 1123-1132.

Mangan PR, Harrington LE, O'Quinn DB, Helms WS, Bullard DC, Elson CO, Hatton, RD, Wahl SM, Schoeb TR, Weaver CT: TGF-β induces development of the Th17 lineage. Nature 2006, in press. This study identified a central role for TGF-β to initiate commitment of naïve CD4 T cells to Th17 development. TGF-β was shown to induce IL-17 expression and to up-regulate the inducible chain of the IL-23 receptor (IL-23R) in the absence of IL-23, placing TGF-β upstream of IL-23 in Th17 development. This study also provided in vivo evidence that links TGF-β to Th17 development, demonstrating that mice deficient in TGF-β1 are deficient in Th17 cells and in circulating levels of IL-17.

TGFβ in the context of an inflammatory cytokine milieu supports de novo differentiation of IL-17-producing T cells
-
This study independently identified a role for TGF-β in Th17 development, showing that natural Tregs could be substituted for TGFβ. This study further showed that proinflammatory cytokines, particularly IL-6, augment IL-17 expression, linking effects of inflammation and natural Treg actions to cooperatively drive Th17 development.
