Arterioscler Thromb Vasc Biol 2004; 24:697-702. This study describes the molecular mechanism employed by LDLs to induce vascular smooth muscle cell proliferation. It shows that pertussis toxin-sensitive GPCR, Ca2+ mobilization, PKC, the ERK and the p38 MAPK pathways are all involved in the upregulation of neuron-derived orphan receptor 1, a transcription factor mediating vascular smooth muscle cell proliferation in response to LDL.

Characterization of the in vivo role of the p38 MAPK pathway in the vascular response to injury. Of particular note is the observation that inhibition of p38 MAPKs abrogates the induction of the proinflammatory cytokine IL-1b and neointima formation in injured arteries. This suggests that the p38 MAPK pathway plays a critical role in the progression of atherosclerosis.
