    • The role of chromosomal instability in tumor initiation
    • The authors developed a mathematical model for studying the effect of CIN on the evolutionary dynamics of colorectal tumors in either the presence or absence of an APC mutation. They calculate the conditions required for CIN to initiate tumorigenesis before the mutation of tumor suppressor genes and suggest that an event creating genetic instability is sufficient to initiate and drive tumor progression in sporadic neoplasias.

    • Identification of Stk6/STK15 as a candidate low-penetrance tumor-susceptibility gene in mouse and human
    • The authors refine the localization of a tumor susceptibility locus to a region of 1-2Mb on chromosome 2 using mouse genetics and further resolve to the level of individual genes by correlative study of an orthologous locus in humans that is preferentially amplified in aneuploid tumors. Their combined strategy using mouse and human models identified the role of a common tumor susceptibility gene, Stk6/STK15, in promoting genetic instability although the mechanisms by which they modify risk differ between mouse and human.
