    • Nucleosome remodeling induced by RNA polymerase II: Loss of the H2A/H2B dimer during transcription
    • This paper describes an experimental system for the study of PolII transcription on a mononucleosomal template. Elongation complexes are assembled with PolII immobilized through the tag and ligated to the mononucleosome. In this system, fully transcribed templates are released into solution and the fate of nucleosomes traversed by PolII can be assessed. The study shows that passage of PolII causes a quantitative loss of one H2A/H2B dimer thus leaving behind an imprint of disrupted chromatin structure. This system allows analyses of the effects of different elongation factors on nucleosome structure during PolII transcription. It was used to demonstrate FACT mechanism in transcription [48].

    • The depletion of either DSIF or NELF both in vitro (immunodepletion) or in vivo (RNAi) results in the reduced level of the promoter proximally paused PolII at the Drosophila HSP70 gene before heat-shock induction. NELF does not co-localize with transcriptionally active genes, whereas DSIF does. The authors propose a model, which would explain both positive and negative roles for DSIF in transcription. Given that DSIF binds to PolII directly, it might function as an adaptor connecting other factors to the elongation complex.

Two major novel observations were made. First, the distribution of the Paf1/Cdc73 complex was found to be similar to that of the known transcript elongation factors, TFIIS and Spt5. This observation suggested a role for the Paf1/Cdc73 complex in transcript elongation. Second, Elongator was not detected in the coding regions of the genes tested and, as was revealed by the immunofluorescence studies, it was mostly localized to the cytoplasm. Thus the role for the Elongator in the PolII transcription apparatus became more obscure.

Chromatin structure of the transcribed genes is altered in spt6 mutants.     • Tracking FACT and the RNA polymerase II elongation complex through chromatin in vivo
    • A combination of results from immunofluorescence staining of Drosophila polytene chromosomes and high-resolution ChIP analyses using antibodies specific to the Drosophila homologues of Spt16 and SSRP1 established that, upon heat-shock induction, FACT is recruited to the actively transcribed loci. The kinetics of FACT recruitment and of chromosome tracking in vivo resembles that of PolII and elongation factors Spt5 and Spt6.
