This technology is used to identify several genes that are synthetically lethal with a mutation involved in double-stranded DNA repair or the chemical camptothecin, thus opening the door to synthetic screening on a scale previously performed only in yeast.

PLoS Biol 2004, in press. This impressive study combines genome-wide RNAi, carried out in triplicate, with testing of 51000 small molecules for their effect on cytokinesis. Very similar phenotypes were elicited by both methods, suggesting that RNAi and small molecule perturbations affect the same pathways. These observations further point to specific genes as potential molecular targets for the activity of small molecules.
