Davis Chemogenomic profiling: identifying the functional interactions of small molecules in yeast Proc Natl Acad Sci USA 101 2004 793 798 In a previous study, the authors determined that heterozygous mutants of drug targets display enhanced sensitivity to the drug. Here, this principle was scaled to the entire genome, using the yeast deletion strain collection. The authors identified potential targets of several drugs by tracking their corresponding deletion strain in mixed populations using genetic barcodes. This report outlines the potential of genome-wide approaches for discoveries outside of normal gene function annotation.

The authors scored their library (10 000 human genes and 5000 mouse genes) for effects on proteasome function, identifying several known members of this class of genes, and thus verifying their approach. In addition, they incorporated multifunctional shRNA cassettes that could be transported to other expression vectors, as well as genetic barcodes, permitting microarray tracking of pooled shRNA collections.

First, known TGFβ signaling components were used as bait to identify interacting proteins. Then, RNAi analysis in both wild-type and TGFβ mutant background worms was used to confirm functional roles of the novel proteins and to establish epistatic relationships between them.
