[30• that localization of Rad51 to nuclear damage foci did not occur in FA-D1 mutant cells, this group has clearly shown that c-terminal truncated BRCA2 alleles missing the c-terminal Rad51 binding domain account for Fanconi anemia of the D1 type. Howlett et al. have successfully tied together the FA and BRCA pathways in an intriguing loop that must be clarified functionally, because the relation between these protein families may have relevance to maintenance of genomic integrity, carcinogenesis, leukemogenesis, and cell survival

By exposing cells from all the known Fanconi anemia complementation groups to ionizing radiation and mitomycin C, these investigators discovered that only in FA-D1 cells did Rad51 fail to localize to nuclear damage foci. They concluded that FANCD1 was likely involved in repair of double-strand breaks by homologous recombination, given the known function of Rad51. These findings also catalyzed the search for a potential role of BRCA2 in FA-D1 cells [12•]

In the author's own scientific life, nothing had been more hotly contested than the arguments on localization and function of the Fanconi anemia proteins. Fortunately, much of the work on the nuclear complex pathway has solidified that the mitomycin C resistance pathway is both linear and nuclear; this pathway, as it is perturbed in all somatic cells, does not account for the unique involvement of hematopoiesis in the Fanconi anemia phenotype. Fortunately, it is now equally clear that some Fanconi anemia proteins have more than one function. The focus of the author's group has been on defining the hematopoietic function of Fanconi anemia proteins. This article describes the creation of site-directed mutations in a conserved domain of FANCC that are capable of fully complementing FA-C cells in mitomycin C assays but that do not complement in signaling assays specific for hematopoietic cells. This advance not only demonstrates the multifunctionality of FANCC but also raises the possibility that mutations of this kind might occur in nature and, if so, that some patients with bone marrow failure resulting from Fanconi anemia gene mutations might have normal mitomycin C tests
