    • Structure of the retinoblastoma tumor-suppressor pocket domain bound to a peptide from HPV E7
    • This paper reveals the crystal structure of the Rb 'pocket' bound to the LxCxE motif from HPV E7. The A/B domain of pRB forms a hydrophobic pocket sustained by critical hydrogen bonds. The B-box contains a cyclin fold which binds the LxCxE motif whereas the A-box ensures the correct structural folding of the B-box. The crystal structure also suggests that a second potentially important protein-binding domain might be formed by the A-B interface.

    • Stable binding to E2F is not required for the retinoblastoma protein to activate transcription, promote differentiation and suppress tumor cell growth

    • Mutant forms of E1A which are defective for binding of either p300 or RB family members demonstrated that both functions of E1A - the ability to bind p300 and RB related proteins - are required for p53 induction and apoptosis in primary cells. When the E1A mutant deficient for binding of RB family members was expressed in Rb-null MEFs, the ability to induce p53 and apoptosis was restored, suggesting that Rb is the key target of E1A in transformation, not p107 or p130.

The findings interestingly reveal a tumor-suppressor function for p107. Basically, mouse retinas are better protected against tumorigenesis than human retinas.
