The antidepressant-like effects induced by desipramine and fluoxetine were similar among wild-type and PDE4D-deficient mice, while the efficacy of rolipram was absent in PDE4D-deficient mice, suggesting that PDE4D plays a critical role in cAMP signaling that is directly linked to mood.

J Physiol (2007) 584(2):401-405. •• Under normal conditions, DISC1 interacts with PDE4B and PDE4B dissociates from DISC1 following increased cellular cAMP levels. Mutations in the DISC1 gene prevent PDE4B binding, while PDE4B mutations result in isoforms that are incompatible with DISC1 regulation. Chromosomal translocation of the PDE4 gene in a patient with schizophrenia resulted in a 50% reduction in PDE4B1 levels and, consequently, increased cellular cAMP levels augmented PDE activity. DISC1 is involved in cAMP signaling and neuronal migration. Disruptions in DISC1 and PDE4B result in alterations in brain development, effect and cognition. The abrogated regulation of cAMP signaling may be directly linked to learning, memory and mood alterations in individuals with clinical depression and schizophrenia.
