Virulence was associated with increased IL-4 production and IgG1 and IgE levels. This provides an explanation for the presence of a multicopy tandem array of CPB genes in L. mexicana.

Furthermore, inserting multicopy L. mexicana CPB genes into L. major decreased the IFN-γ response and increased parasite numbers in C3H mice; thus, CPB is a determinant of increased virulence of L. mexicana, by comparison with L. major, for most mouse strains.

TGF-β has been shown not only to downregulate cellular immunity against Kala-azar but to promote survival within the host macrophage. Leishmanial cysteine peptidases and, in particular, cathepsin B (CPC) were implicated in the activation mechanism of TGF-β.
