J Biol Chem 1997, 272:14285-14293. In this paper six of the 19 cpb genes were analysed for sequence variation and expression profiles in different life-cycle stages. Genes 1 and 2, which lack the characteristic carboxy-terminal extension, were expressed in the infective metacyclic stage, whereas the remaining genes in the array, which contain the carboxy-terminal extension, were amastigote specific. Individual isoenzymes re-expressed in a cpb null mutant were found to vary in substrate specificity and ability to restore loss of virulence associated with the null mutant.

T. cruzi epimastigotes, when transfected with cosmid clones containing about 20 copies of the cruzipain gene, were found to over-express cysteine proteinase activity. This upregulation was maintained throughout the life-cycle with the enzyme found in vesicles of the endosomal/lysosomal system in epimastigotes, but also detected on the cell surface of the amastigote. Phenotypic analysis showed that the transformed parasites had an enhanced ability to undergo metacyclogenesis.

The major cysteine proteinase of T. cruzi, cruzipain, isolated and purified from the parasite was found to cleave kininogen to generate Lys-bradykinin, a proinflammatory peptide. Interestingly, two recombinant cruzipain isoenzymes, cruzain and cruzipain 2, cleaved the bradykinin-flanking sites at markedly different rates. Neither of the two recombinant enzymes contain the characteristic carboxy-terminal extension and the authors tentatively propose that this domain may influence the enzymatic properties of cruzipain. It should be noted, however, that the enzyme purified from the parasite contains multiple isoenzymes derived from up to 40 gene copies. Differences in substrate specificity may therefore arise from isoenzymes transcribed from different copies within the cruzipain gene array.

A model of the L. major cathespin L- and cathepsin B-like cysteine proteinases was constructed on the basis of the known crystal structure of other papain-family enzymes. Potential cysteine proteinase inhibitors were identified by computer searching using the program Dock3.5. Several compounds selected from this screen were found to inhibit greatly at concentrations as low as 50 μM. In a subsequent study these inhibitors were found to have potent anti-leishmanial activity both in vitro and in vivo (PM Selzer et al., personal communication).

Treatment of T. cruzi with cysteine proteinase-specific inhibitors arrested cell growth and altered the intracellular location of cruzain by causing a build up in the Golgi compartment. The results suggest that the proteinase inhibitors prevent auto-catalytic processing of the enzyme resulting in an accumulation in the Golgi and causing a peripheral distention of the cisternae. This build up of pre-cursor parallels death of the epimastigote.

This study confirmed that leishmanial cysteine proteinases can induce a Th1-type immune response but importantly also showed that vaccinating BALB/c mice with the enzyme can provide protection against a subsequent challenge with L. amazonensis amastigotes.
