    • + T cells. Increased IL-4, IL-10 and IFN-γ production in ICOS costimulated T cells is associated with strong activation of the PI3-K-PDK1-Akt/PKB pathway. In addition the MAPK family kinases are activated in different ways by ICOS and CD28. Interestingly, the natural skewing towards Th2 cytokine responses in Balb/c versus Th1 dominated cytokine responses in C57BI/6 mice is associated with a much higher expression of ICOS on Balb/c versus C57BI/6 CD4+ T cells.

Inducible costimulatory molecule-B7-related protein 1 interactions are important for the clonal expansion and B cell helper functions of naive, Th1, and Th2 T cells. J Immunol 2003; 170:2310-2315. Furthermore, although it has no effect on the migration of T cells into B cell follicles, it is essential for their ability to support B cell responses.

Nat Med 2002; 8:1024-1032. This paper claims to induce regulatory T cell development after respiratory exposure to allergen (ovalbumin) in the presence of IL-10 producing mature pulmonary dendritic cells. The authors provide evidence that these cells ameloriated ovalbumin induced asthma IL-10 and ICOS dependent pathway.

In this study, the role of ICOS in NK cell mediated cytotoxicity and cytokine production was assessed. To determine whether ICOS contributes to the elimination of tumors in vivo, tumor growth and the survival of mice injected with the tumor expressing ICOS-L were examined. So far this is the only paper demonstrating effects of ICOS costimulation in NK cell activity and its potential importance in tumor surveillance.
