PD-1 immunoreceptor inhibits B cell receptor-mediated signaling by recruiting src homology 2-domain-containing tyrosine phosphatase 2 to phosphotyrosine. Proc Natl Acad Sci USA. 98:2001;13866-13871. In this article, PD-1 is shown to inhibit B cell activation by competing B cell receptor signaling. PD-1 is shown to recruit SHP-2 upon coligation with B cell receptor and dephosphorylate key molecules in B cell activation. Thus, this article provides the first molecular basis for inhibitory effects of PD-1 on activation of lymphocytes.

Engagement of the PD-1 immunoinhibitory receptor by a novel B7 family member leads to negative regulation of lymphocyte activation. J Exp Med. 192:2000;1027-1034. Together with [18•], these articles describe identification of ligands of PD-1, PD-L1 and PD-L2. These ligands belong to the B7 family and inhibit T cell proliferation and cytokine production in vitro.

Science. 291:2001;319-322. PD1 deficient mice on the BALB/c background are shown to develop fatal dilated cardiomyopathy. This disease involves high-titered heart specific IgG autoantibodies in sera and their deposition on the surface of cardiomyocytes. This is the first animal model for the dilated cardiomyopathy caused by autoimmunity.

    • Rottman J.B., Smith T., Tonra J.R., Ganley K., Bloom T., Silva R., Pierce B., Gutierrez-Ramos J.C., Ozkaynak E., Coyle A.J. The costimulatory molecule ICOS plays an important role in the immunopathogenesis of EAE. Nat Immunol. 2:2001;605-611. ICOS blockade is shown to either protect or facilitate EAE, depending on the timing of blockade. ICOS blockade at the induction phase of EAE exacerbates the disease and ICOS blockade at the effector phase abrogates the disease.
