Genes Dev 1998, 12:3663-3674. The authors provide evidence that c-Cbl sorts EGFR in the endosome compartment for degradation and that oncogenic mutants of Cbl interfere to favor recycling of EGFR. Interference with a genetically defined negative regulator of EGFR signaling results in enhanced mitogenesis due to altered endocytic trafficking.

EMBO J 1998, 17:554-564. The hdm2 oncoprotein shuttles between the nucleus and the cytoplasm and its export is blocked by inhibitors of Rev export. The export function of hdm2 is required to accelerate p53 degradation and blockade of the Rev pathway inhibits negative regulation of hdm2 on p53 transcriptional activity.

Mdm2 binds to NUMB and causes translocation of the latter to the nucleus. This event is associated with a reduction in the levels of NUMB. Degradation of NUMB might be part of the oncogenic action of mdm2.

Loss of TSC2 causes instability of the p27 negative growth regulator and resistance to the activity of ectopic p27. In TSC2-negative cells p27 is mislocalized to the cytosol where it might be degraded.
