Methods used for vaccinia infection, cell culture, and FcεRl stimulation were reported (5). Briefly, RBL-2H3 cells were infected with five plaque forming units (PFU) per cell of recombinant porcine Syk vaccinia and the indicated dose of recombinant wild-type Cbl, 70Z Cbl, 1-480 Cbl, 1-655 Cbl, or vaccinia vector in Eagle's minimum essential medium (EMEM) with 5% fetal bovine serum for 2 hours, washed once with EMEM, and incubated for an additional 6 to 8 hours in EMEM with 5% fetal bovine serum and monoclonal anti-lgE to DNP (1 ng/ml). Infected cells were harvested with phosphate-buffered saline containing 1 mM EDTA, washed with EMEM and resuspended in EMEM. Cells were stimulated with DNP-HSA (200 ng/ml) for 2 min or the indicated time.
