Biochemical evidence that RhoB is a target of FPTase inhibitors. RhoB is both farnesylated and geranylgeranylated, and the results of this paper show that an FPTase inhibitor blocks RhoB farnesylation in cells. Since RhoB was previously shown to be required for Ras function, the results suggest that inhibition of RhoB farnesylation by FPTase inhibitors may be a mechanism by which these compounds exhibit an anti-proliferative effect.
