Genetic inhibition of the protease inhibited Ras function in yeast but was not toxic to the yeast. This work should lead to the cloning of mammalian CaaX protease, which is a potential target for the development of inhibitors with anti-Ras activity.

Since this GTP-binding protein was found to inhibit Ras functions, alteration of Rheb activity might also affect Ras pathways.

Biochemical evidence that RhoB is a target of FPTase inhibitors. RhoB is both farnesylated and geranylgeranylated, and the results of this paper show that an FPTase inhibitor blocks RhoB farnesylation in cells. Since RhoB was previously shown to be required for Ras function, the results suggest that inhibition of RhoB farnesylation by FPTase inhibitors may be a mechanism by which these compounds exhibit an anti-proliferative effect.

This result raised the possibility that FPTase inhibitors may not effectively inhibit the growth of Ki4B-ras transformed cells and that any inhibition observed might be due to inhibition of other farnesylated proteins that regulate cell proliferation.
