Burgstahler AD, Nathanson MH: NO modulates the apicolateral • cytoskeleton of isolated hepatocytes by a PKC-dependent, cGMP-independent mechanism. Am J Physiol 1995, 269:G789-G799. Although NO induces smooth muscle relaxation, in isolated hepatocyte couplets NO was shown to potentiate rather than inhibit apical cytoskeletal contraction. This stimulation occured via activation of protein kinase C, rather than by a cGMP mechanism. These observations suggest a new role for NO: to activate protein kinase C.

Am J Physiol 1996, 271:G293-G303. The signaling mechanisms for bile acid regulation of the rate-limiting enzyme for bile acid biosynthesis from cholesterol, cholesterol 7α-hydroxylase, were examined. Bile acids were found to activate intracellular protein kinase C, resulting in its sequential redistribution and downregulation of both calcium-dependent and -independent isoforms of this kinase. This study indicates that bile acids can act as ligands to stimulate intracellular signaling pathways for nuclear gene transcriptional regulation.

This group has previously shown that apical plasma membrane proteins must route through the basolateral plasma membrane before retrieval and delivery to the apical pole of the hepatocyte. In this study, dipeptidyl peptidase IV is shown to be endocytosed from the basolateral membrane in vesicles that are distinct from the clathrin-coated vesicles containing polymeric IgA receptor. Transcytosis of these two proteins from basolateral early endosomes to the subapical region appears to occur via a single transcytotic pathway. The mechanisms for sorting and regulation of the transcytotic pathway are under continued investigation.

J Biol Chem 1996, 271:2341-2346. The Gunn rat, deficient in the hepatocellular enzyme necessary for bilirubin conjugation, was used to evaluate gene-targeting to the liver. The appropriate DNA was complexed with galactose-terminated (asialo-) glycoprotein-polylysine conjugates, so as to take advantage of receptor-mediated endocytosis of the asialoglycoprotein. Disruption of hepatocellular microtubule function with colchicine led to marked persistence of the DNA in the liver, suggesting a new noninvasive method for prolonging the effect of liver-targeted gene therapy.

Dynamins are a family of 100-kd GTPases that are thought to facilitate the formation of endocytic vesicles, and hence promote intracellular vesicular traffic. This study documents that a dynamin-like protein associates with the Golgi apparatus and may therefore play a role in remodeling and trafficking of vesicles in multiple sites within the cell. The role of these proteins in hepatocyte vesicular trafficking is under investigation.

Unesterified cholesterol exchanges between circulating lipoproteins and the hepatocyte basolateral plasma membrane, whereas cholesteryl esters can only be liberated after hepatocellular endocytosis and degradation of lipoproteins. The plant sterol sitostanol can be used as a nonmetabolizable analogue of free cholesterol. Sitostanol, incorporated into phosphatidylcholine liposomes, was taken up from the perfusate of isolated rat livers and appeared rapidly in bile; biliary secretion was dependent on bile salt secretion. The rapid transit from plasma-to-bile paralleled that of free cholesterol. These studies suggest that sitostanol can be used as a physiologic analogue of unesterified cholesterol to trace the transport of sterols through the liver.

In addressing the trafficking of small amphipathic organic molecules through the liver, most attention focuses on the function and molecular regulation of protein transporters in the basolateral and canalicular plasma membrane. This review addresses research demonstrating that diffusion of these molecules through the cytoplasm also may constitute a rate-limiting process for cellular transport and metabolism. Moreover, inherited storage diseases may seriously disrupt normal cytoplasmic transport mechanisms.

This finding supports the concept of hepatocytes acting as immunomodulatory cells.

Leukocyte trafficking through the hepatic microvasculature was studied with intravital fluorescence microscopy, using a model of ischemia-reperfusion. Hepatic reperfusion after ischemia was characterized by perfusion failure of individual sinusoids and a significant increase in sinusoidal leukostasis. However, most sinusoids with stagnant leukocytes were still perfused. Thus, sinusoidal leukostasis per se does not appear to be sufficient to induce perfusion failure ("no-reflow") after ischemia-reperfusion of the liver.

A differential polymerase chain reaction technique was used to demonstrate that perisinusoidal stellate (Ito) cells increase their expression of ICAM-1 following 1 week of bile duct ligation. Thus, the stellate cells may have a direct role in the transmigration of neutrophils from the hepatic sinusoid during inflammation.

The mechanism of ethanol-induced hepatic inflammation was examined by determining whether short-term ethanol exposure stimulated production of rat interleukin-8 (CINC) by hepatocytes or Kupffer cells. There was no direct stimulation by ethanol, but cytokines and endotoxin induced CINC production by both cell types. as did conditioned medium from ethanol-pretreated hepatocytes. The mechanism of ethanol-induced neutrophil infiltration into the liver is therefore likely to be indirect, involving ethanol oxidation and communication between hepatocytes and Kupffer cells.

Sodium-dependent basolateral bile acid transport was greatly reduced, as was ATP-dependent and membrane potential-dependent canalicular transport The mRNA levels and immunoblot antigenic content of the putative membrane transporters were similarly decreased. Impaired bile acid transport at the sinusoidal and canalicular membrane domains by these and other mediators of the inflammatory response may account for sepsis-associated cholestasis.

Hepatology 1996, 23:797-802. Stimulation of NO release by hepatocytes and Kupffer cells was examined. Interleukin-1β alone was capable of stimulating NO release by hepatocytes. TNF-α and interleukin-6 were capable of modulating the interleukin-1β effect but could not stimulate hepatocytes directly. None of the cytokines tested stimulated NO production by Kupffer cells. This study demonstrates that some cytokines may have direct and immediate effects on hepatocytes.

Cholangitis was induced in rats by immunization with highly purified bile duct epithelial cells (cholangiocytes). Recipient animals developed histological evidence of nonsuppurative cholangitis without inflammation in other organs. Cholangiograms showed distortion and tortuosity of the entire intrahepatic biliary ductal system. This unique rodent model of experimental cholangitis may prove useful in exploring the mechanisms by which the immune system targets and damages cholangiocytes.
