Cyster Lymphocyte egress from thymus and peripheral lymphoid organs is dependent on S1P receptor 1 Nature 427 2004 355 360 Using mice whose haematopoietic cells lack a particular S1P receptor (S1P1) the authors demonstrate that S1P1 is essential for lymphocyte recirculation, and that it regulates egress from both thymus and peripheral lymphoid organs. Importantly, S1P1-dependent chemotactic responsiveness is strongly upregulated in T cell development before exit from the thymus, whereas S1P1 is downregulated during peripheral lymphocyte activation, and this is associated with retention in lymphoid organs.

This agonist established that the effect of S1P on lymphocyte recirculation occurs through S1P1 and not S1P3. An undesired effect of broad S1P receptor agonists is bradycardia, and this was shown to be abolished in S1P3-deficient mice. S1P1 appears to be an attractive target that could be used to interfere with lymphocyte trafficking.
