    • Plasma cell differentiation and the unfolded protein response intersect at the transcription factor XBP-1
    • This study nicely demonstrates that the expression of an XBP-1 splicing variant is linked with full plasma cells commitment but also importantly with a very unique capacity of plasma cells to handle the enormously enhanced metabolic/mitochondrial stress induced by immunoglobulin production.

    • Initiation of plasma-cell differentiation is independent of the transcription factor Blimp-1
    • -/- mice reconstituted with Prdm1 mutant fetal liver lack a defined plasma-cell compartment. However, detectable amounts of immunoglobulin were found. As a result, Blimp-1-independent differentiation occurs at a preplasmablast level, and early stages before Blimp-1 expression appeared to be initialized by Pax5 inhibition. These data support that plasmablast/plasma-cell subsets follow a specific differentiation path that is under control of distinct transcription factors. How this is clearly linked to phenotypic characteristics is unknown.

    • This report contains beautiful in vivo imaging of germinal center dynamics, clarifying many aspects of the cellular dynamics of this process. Also highlighted the possibly key role of the T cell B cell interaction as the mediator of selection that drives affinity maturation. That is, B cells may be competing for access to limited T cell help rather than for limiting antigen on follicular dendritic cells.

    • This paper describes the other half of Bcl6's activities in the germinal center, showing how it suppresses aspects of the DNA damage response. Interesting also in that this repression is relieved by CD40 stimulation, consistent with a cyclic reappraisal of a centroblasts progress at around the same time at which choices of differentiation may also be being made.
