This seminal study describes KO mice lacking T1R1, T1R2, and T1R3, which demonstrates that T1R1+3 is essential for amino acid taste and T1R2+3 is essential for sweet taste in vivo. Replacing the mouse T1R2 gene with the human T1R2 gene produces mice with humanized sweet taste preferences, which suggests that variations in T1R receptors account for species-specific taste preferences. Introducing a GPCR that is activated by an artificial ligand into T1R2-containing cells results in the mice preferring the artificial ligand, which shows that taste cells are hardwired to stereotyped behaviors.

This is a landmark study that identifies signaling components necessary for sweet and bitter taste detection and in doing so eliminates a plethora of proposed taste transduction cascades. Knockout mice lacking PLCβ2 or TRPM5 do not detect sugars or bitter substances, which demonstrates that taste detection is mediated by a PLC-activated cascade and that no other signaling cascades act independently.
