    • Putative mammalian taste receptors: A class of taste-specific GPCRs with distinct topographic selectivity
    • T1R1 and T1R2 are two putative taste receptors selectively expressed in TRCs. Like the calcium-sensing receptors, vomeronasal receptors and mGluRs that they are most closely related to, T1R1 and T1R2 are predicted to have a large amino-terminal extracellular domain that may participate in ligand binding. To date, however, there are no functional data on their ligand-binding properties. The topographic pattern of expression of T1R1 and T1R2 in TRCs does not match the regional sensitivity of any tastants.

    • T2Rs function as bitter taste receptors
    • Using heterologous expression of chimeric T2R/TRB receptors containing the amino terminus of rhodopsin, it was demonstrated that cells transfected with chimeric mT2R-5 respond only to the bitter antibiotic cycloheximide (from among 55 bitter and sweet compounds tested). Five amino-acid differences were noted between mT2R-5 from cycloheximide-sensitive versus insensitive strains of mice: it was proposed that these sequence differences underlie the in vivo differences between the taster and nontaster strains. mT2R-5 selectively couples to α-gustducin over any other α-subunit tested, which is appropriate given the colocalization of gustducin and the T2R/TRB receptors and gustducin's demonstrated involvement in bitter taste transduction. The authors have convincingly demonstrated that mT2R-5 is a bona fide taste receptor, suggesting that the other members of this family may also be bitter receptors.

A depolarizing response to glutamate (mimicked by NMDA and inhibited by NMDA antagonists) is suggested to represent synaptic glutamate receptor activity. Hyperpolarizing responses are also seen that are mimicked by the metabotropic receptor agonist, L-AP4, and antagonized by cAMP. This response has the properties predicted for a current activated by the putative umami receptor, taste-mGluR4.

    • A truncated variant of mGluR4, designated taste-mGluR4, is cloned from taste tissue and shown to be selectively expressed in taste tissue. Functional expression of taste-mGluR4 in CHO cells demonstrates that this receptor has reduced sensitivity to L-glutamate than does brain-expressed mGluR4, that is similar to the umami taste receptor. The effects of L-Glu on the cloned and expressed taste-mGluR4 are mimicked by the mGluR4 agonist, L-AP4, which elicits a similar taste response to monosodium glutamate. In CHP cells, the expressed receptor is shown to be negatively coupled to cAMP production, suggesting possible involvement of a cyclic-nucleotide-gated channel in the umami response. The results are consistent with the taste-mGluR4 being the transduction channel for glutamate taste.
