    • Further proof of the central role of the DCL4 pathway in mediating RNAi induced by hairpin RNA. This paper includes much data of relevance to biotechnologists, including compelling evidence that hairpin RNAs can also produce siRNAs via the DCL2 and DCL3 pathways. It is likely that some of the unexpected and poorly explained problems with RNAi are due to this relative lack of specificity.

As explained in the main text, miRNA precursors are processed in a much more predictable and specific manner than long hairpin RNAs, reducing the potential for unexpected effects. A systematic study of a large number of amiRNAs needs to be completed to discover if the theoretical advantages can be fully realised in practice.

    • A superb demonstration of the potential of RNAi in plant biotechnology. Gossypol toxin, normally found throughout the plant, improves pest tolerance of cotton but makes the oil extracted from seeds inedible. By specifically reducing expression of δ-cadinene synthase in developing seeds using tissue-specific RNAi, the authors succeeded in producing gossypol-free seeds whilst maintaining normal levels of the toxin elsewhere in the plant. If this approach turns out to be stable in the field and passes regulatory hurdles, it could considerably increase the value of cottonseed oil.
