Smad7 in epidermis associates with β-catenin, after which β-catenin is degraded via recruitment of Smurf2 to the complex. Double transgenic mice for Smurf2 and Smad7 in epidermis aggravates the abnormality seen in Smad7 transgenic mouse, and knock-down of Smad7 increases Wnt signaling in keratinocytes. Thus, Smad7 antagonizes Wnt/β-catenin signaling to alter skin differentiation program.
