    • This article reviews recent progress in the simulation of protein folding kinetics and thermodynamics, and protein structure prediction, with an emphasis on quantitative comparison with experiments.     • Solution structure of a protein denatured state and folding intermediate
    • The role of intermediate states in protein folding remains a controversial subject. The authors provide evidence that the denatured state of EnHD is a productive folding intermediate. Using NMR methods, they solve the high-resolution structure of a mutant of EnHD (L16A) at physiological ionic strength, where it populates the intermediate state. The structure shows that the helices are formed in the intermediate state and poised to dock with one another to form the native structure.

    • This work represents an effort to engineer the folding of an ultrafast folding protein towards the folding speed limit. VHP was mutated to remove unfavorable electrostatic interactions in the native structure, thereby decreasing the folding lifetime to less than 1 μs. Despite being the fastest folding protein discovered to date, this mutant still has a small folding activation barrier.
