The authors examined the location of several species of Aβ and observed significant intracellular Aβ1-42 immunohistochemistry in the temporal cortex from 70 individuals ranging in age from 3 to 73 years. In addition, more extensive intracellular Aβ1-42 was observed in young individuals (3-4 years) but decreased as extracellular Aβ1-42 increased. The authors also provide evidence that intracellular Aβ could be clearly distinguished from intracellular APP.

These abnormalities can be paralleled in astrocyte cultures by the inhibition of mitochondrial metabolism. Early abnormal APP processing may thus be caused by mitochondrial dysfunction in DS.
