Wild-type and CD14-deficient mice were injected with E. coli LPS. Wild-type mice had an increase in myocardial NF-κB activation, cytokine mRNA production, and hemodynamic parameters showing depressed left ventricular function. CD14-deficient mice, on the other hand, had lower levels of NF-κB activation and cytokine mRNA production, and also retained normal reft ventricular function. These findings strongly suggest that the TLR4-CD14 pathway is involved in the pathophysiology of depressed cardiac function seen in sepsis.

In the absence of MD-2, TLR4 was confined to the Golgi, indicating that MD-2 plays a role in the intracellular distribution of TLR4 in the LPS response.
