    • Leptin activates anorexigenic POMC neurons through a neural network in the arcuate nucleus
    • This study used an electrophysiological approach to characterise responses to leptin in pro-opiomelanocortin (POMC) neurones identified by selective expression of green fluorescent protein in transgenic mice. The authors were able to demonstrate activation of these cells by leptin, reduced inhibition from local neuropeptide Y and GABAergic neurones and an autoinhibitory effect of melanocortins.

    • Ciliary neurotrophic factor activates leptin-like pathways and reduces body fat, without cachexia or rebound weight gain, even in leptin-resistant obesity
    • Ciliary neurotrophic factor does not play a physiological role in weight regulation but activates similar post-receptor signaling pathways to leptin and appears to be more effective at reducing weight than leptin in animal models. Trials in human obesity are underway.
