This study used an electrophysiological approach to characterise responses to leptin in pro-opiomelanocortin (POMC) neurones identified by selective expression of green fluorescent protein in transgenic mice. The authors were able to demonstrate activation of these cells by leptin, reduced inhibition from local neuropeptide Y and GABAergic neurones and an autoinhibitory effect of melanocortins.

AZIPF1 lipoatrophic mice are characterised by having virtually no fat, are markedly insulin resistant and have severe hepatic steatosis. This paper demonstrates that leptin replacement can improve insulin resistance and glucose metabolism in this animal model of lipoatrophic diabetes and could lead to trials of leptin treatment in humans with lipodystrophy.
