    • Directed evolution of new catalytic activity using the alpha/beta-barrel scaffold
    • An existing binding scaffold was endowed with catalytic activity by designed alterations followed by DNA shuffling. This resulted in the conversion of indole-3-glycerol-phosphate synthase to an efficient phosphoribosylanthranilate isomerase. Designing a binding site is easier than designing a catalytic site and, in this case at least, the results indicate that evolution can convert the former to the latter. This is important as it has implications on the choice of starting point in evolution experiments where a new catalytic activity is sought.
