    • This study and [7] demonstrate the essential role of sphingolipids in the formation of Triton X-100-insoluble lipid domains, which are greatly enhanced by the presence of cholesterol. Moreover it shows that although Triton X-100 insolubility characterizes the liquid-ordered (lo) relative to the liquid-disordered (ld) phase, this insolubility appears to significantly underestimate the amount of lo phase present. Detergent does not, however, artifactually create ordered domains or induce components of solubilized membranes to associate with detergent-insoluble membranes in separate bilayers within the same lysate

In this paper, the authors also show that crosslinking of FcεRI induces the co-aggregation of membrane rafts - as indicated by the co-redistribution of Lyn and GD1b at the cell surface - and that only Lyn co-redistribution is prevented by cholesterol depletion, consistent with the biochemical data

    • LAT palmitoylation: Its essential role in membrane microdomain targeting and tyrosine phosphorylation during T cell activation
    • This is the first demonstration of LAT palmitoylation on the two juxtamembrane cysteine residues, Cys26 and Cys29. Although both cysteine residues contribute to LAT palmitoylation, Cys26 is essential for the association of LAT with rafts. Furthermore this paper, together with [34,35], shows that mutations at Cys26 and Cys29 or Cys26 alone, but not Cys29 alone, abolish LAT function and TCR signaling. In [35], the authors show, in addition, that the extracellular and transmembrane domains of LAT are dispensable for TCR-mediated signaling events once LAT has localized to membrane rafts
